Semi-synthetic Antibody Library Construction
Semi-synthetic antibody library is an antibody library composed of a part of synthetic variable region sequences and another part of natural sequences. The main purpose of constructing a semi synthetic antibody library is to optimize the sequence of antibodies. The construction of such antibody libraries is mainly to use the heavy chain, light chain or rearranged variable region fragments of the germline, and one or more CDRs are randomly rearranged. The CDR region of the antibody can be partially or completely randomized by direct oligonucleotide mutation or PCR based methods, including CDR3 sequence randomization, CDR3 length randomization, and all CDR regions randomization.
